Research Project
After graduating from the Biochemistry program at the University of Guelph, I have re-joined the lab for another two semesters. During this time I will be examining the changes in gene expression in human hepatoma cells (HepG2) due to treatment with 2,3,7,8-tetrachlorodibenzo-p-dioxin (TCDD), across both time and dose. I will also be examining the primary transcriptional effects of TCDD in mouse hepatocytes (Hepa1c1c7 wild type) by co-treating the cells with cycloheximide, an inhibitor of translation. The gene expression profiles will be examined after one hour and four hours of treatment with TCDD, with a one hour pre-treatment with cycloheximide, to study both the early and intermediate transcriptional effects of TCDD.
Besides the lab work I will also be performing the statistical analyses on the microarrays generated from these studies, including the quality control/quality assurance procedure as well as the normalization, probabilistic and correlation analyses.
Other projects include verifying microarray data by quantitative real-time PCR that Cora Fong generated in a study examining the effects of dimethylsulfoxide (DMSO) in serum-free media on mouse hepatocytes.
