UDP/dTDP-D-glucosyltranferase GtfB from

the chloroeremomycin biosynthetic pathway

A stereo ribbon drawing of UDP/dTDP-D-glucosyltranferase GtfB from the chloroeremomycin biosynthetic pathway.  This family of bi-domain enzymes is constructed of two Rossmann folds (with the beta sheets shown in blue).  The N-terminal domain (bottom) binds the vancomycin heptapeptide aglycone, while the C-terminal domain binds UDP-glucose.  GtfB transfers the glucose from UDP-glucose to a hydroxyl on the 4th residue of the vancomycin heptapeptide aglycone.

dTDP-L- epi-vancosamsyltranferase GtfA from

the chloroeremomycin biosynthetic pathway

A stereo ribbon drawing of dTDP-L-epi-vancosaminyltranferase GtfA from the chloroeremomycin biosynthetic pathway; the beta sheets of the two Rossmann folds are shown in green.  The N-terminal domain has the monoglucosylated heptapeptide acceptor bound, while the C-terminal domain has bound dTDP.  GtfA transfers L-epi-vancosamine from dTDP-L-epi-vancosamine to a hydroxyl on the 6th residue of the vancomycin heptapeptide aglycone.

dTDP-L-vancosaminyltranferase GtfD from

the vancomycin biosynthetic pathway

A stereo ribbon drawing of dTDP-L-vancosaminyltranferase GtfD from the vancomycin biosynthetic pathway; the beta sheets of the two Rossmann folds are shown in magenta.  The N-terminal domain has the monoglucosylated heptapeptide acceptor bound, while the C-terminal domain has bound dTDP.  GtfD transfers L-vancosamine from dTDP-L-vancosamine to the 2’ hydroxyl on the glucose of the monoglucosylated heptapeptide acceptor.
